Leqvio 284 mg per 1.5 ml contains inclisiran, a prescription cholesterol lowering medicine given by subcutaneous injection. It is used with diet in adults with primary hypercholesterolaemia, including heterozygous familial hypercholesterolaemia, or mixed dyslipidaemia when additional LDL cholesterol reduction is required.

Approved uses:
• Use with the maximum tolerated statin dose, with or without other lipid lowering treatment, when LDL cholesterol remains above the agreed target.
• Use alone or with other lipid lowering treatment when statins are not tolerated or are contraindicated.
• Long interval LDL cholesterol management as part of continued diet, medication and laboratory follow up.

How it works:
Inclisiran is a small interfering RNA that reduces liver production of PCSK9. This increases recycling of LDL receptors on liver cells and removes more LDL cholesterol from the blood. It lowers LDL cholesterol, but patients should not be told that it independently prevents heart attack or stroke because direct cardiovascular outcome evidence is not yet established in the approved information.

Active ingredient and composition:
Each prefilled syringe contains inclisiran sodium equivalent to inclisiran 284 mg in 1.5 ml, equal to 189 mg per ml. Excipients are water for injections, sodium hydroxide and concentrated phosphoric acid for pH adjustment.

How to use:
A healthcare professional administers one 284 mg subcutaneous injection initially, a second dose after three months, then one dose every six months. It is usually injected into the abdomen, upper arm or thigh. Continue the prescribed statin or other lipid lowering therapy and diet unless the clinician changes the plan. Missed doses require rescheduling according to the leaflet and clinical team.

Important safety information:
Not for self injection. Discuss previous allergy, severe liver disease, pregnancy and breastfeeding. Use during pregnancy is preferably avoided. Injection should be postponed or reviewed if the syringe is damaged or the solution contains visible particles. Continue cholesterol testing and cardiovascular risk management because Leqvio complements rather than replaces diet and other prescribed therapy.
